    Abstract:近年来,随着人们生活水平的不断提高,对信息的需求呈现出多样化的发展态势。电视台新闻节目同样也受到了各类新媒体的影响,在当前的融媒体时代发展进程中迎来了众多挑战,但另一方面,电视新闻节目迎来的也有创新式发展机遇。基于此,本文从媒介融合和融媒体的实质特点入手,深入探究当前我国电视新闻节目传播中所迎来的挑战和机遇,并对融媒体时代电视新闻节目传播路径与策略进行深入的探究,希望能够对我国当前电视新闻节目发展起到一定的参考和借鉴。

  • Study on the Library Space Reconstruction Based on Reader's Attribute Analysis

    Abstract: [Purpose/significance] At present, many libraries have experienced the phenomenon of insufficient space and environmental aging. Space reconstruction has become an urgent problem to be solved in the library.[Method/process] sing the principle of five elements of library space requirements, the analysis of library space shortage from the perspective of readers' attributes shows that the reason is readers' different needs for library space and grasping the characteristics of readers' attributes makes better space reconstruction.[Result/conclusion] The ideas based on different types of libraries will effectively improve the situation of insufficient libraries space or high reading space vacancy rate.

  • Key Technology and Trends of the Next Generation Repositories

    Abstract: [Purpose/significance] By investigating the research status and service demand of the new generation knowledge at home and abroad, this paper analyzes the key technologies and functional characteristics of the next generation repositories. It puts forward the key trends of institutional repositories, and provides some suggestions for the development of next generation repositories.[Method/process] Through literature research, this paper summarized the development trend of institutional repository technology and function. Then, it introduced 11 key technologies, standards and protocols of the new generation repositories. By researching and designing the functions, it analyzed the development of next generation repositories in many aspects such as framework, functions and service target.[Result/conclusion] The paper generalizes the development trends of next generation repositories as followings:①from institutional repository to institutional information infrastructure.②from self-archiving to automated submission or ingestion.③ from independent platforms to current research information system(CRIS).④ from scholarly outputs management platform to academic resources service center. ⑤from information retrieval to supporting big data and semantic retrieval. ⑥from academic archives to academic scholarly communities. ⑦from providing metrics and altmetrics services to creating a new system of academic evaluation.

  • EFFECT OF Mo CONTENT ON THE MICROSTRUCTURE AND PROPERTIES OF CrMoN COMPOSITE COATINGS

    Abstract: Ceramic coatings are usually used as protective coatings to improve performance and durability of tools and components now. Compared with conventional TiN based hard coating, CrN based coating like Cr-X-N (X=Ti, Al, Si, C, B, Ta, Nb, Ni) is a more interesting choice because of low friction coefficient, superior oxidation resistance and excellent corrosion resistance under severe environment conditions. The CrMoN is among these coatings and attractive since self-lubricating phase MoO3may be formed in tribological process. However the effect of Mo content on structure and tribological properties of CrMoN coatings is not still clear. In the present study, CrMoN composite coatings with different Mo content were deposited on M2 high speed steel (HSS) substrates by DC reactive magnetron sputtering. The effect of Mo content on the microstructure and properties was investigated systematically, including the chemical composition, phase structure, chemical valence, cross-section morphologies, microhardness and tribological properties. The results showed that the phase transformation of the as- deposited coatings occurred with the increase of Mo content. The phase structure changed to (Cr, Mo)N substitutional solid solution based on CrN-type firstly, and then to mixed phase with g-Mo2N as main phase, and a small amount of elemental bcc-Mo phase appeared when the Mo content is 69.3%. The microhardness of the CrMoN composite coatings always increased until the highest hardness when the Mo content reached to 45.4%, and then decreased; a relatively low friction coefficient was obtained compared with that of the CrN coating when more than 45.4%Mo content was doped. The reason is that the more MoO3 lubricant phase could be formed in tribological process.

  • 共培养体系中猪骨骼肌卫星细胞对肌内前体脂肪细胞脂质沉积的影响

    Abstract:本试验旨在研究共培养体系中猪骨骼肌卫星细胞对肌内前体脂肪细胞脂质沉积的影响。试验分离培养了猪骨骼肌卫星细胞和肌内前体脂肪细胞,2种细胞分离培养鉴定后,接种于Transwell细胞小室共培养板进行共培养,待细胞密度达到90%以上分别进行诱导分化。骨骼肌卫星细胞诱导分化8 d后,检测肌内前体脂肪细胞分化水平、分化标志基因的表达以及脂质代谢关键酶的表达。结果显示:与肌内前体脂肪细胞单独培养相比,共培养体系内肌内前体脂肪细胞的脂滴数量和脂滴面积极显著减少(P<0.01),肌内前体脂肪细胞增殖分化转录因子过氧化物酶体增殖物激活受体γ和CCAAT增强子结合蛋白的基因和蛋白表达水平极显著下降(P<0.01),肌内前体脂肪细胞的乙酰辅酶A羧化酶、脂肪酸合成酶的基因和蛋白表达水平极显著下降(P<0.01)。

  • M145F/F146M突变对光受体蛋白细菌视紫红质和古紫质4光循环的影响

    Abstract:古紫质4(archaerhodopsin 4, aR4)与细菌视紫质(bacteriorhodopsin, bR)同属于盐杆菌科,同源性59%,均为光驱质子泵。其功能是在光照条件下将质子由胞内泵到胞外形成跨膜质子梯度,该梯度差被膜上另外一种蛋白ATP合成酶用于ATP的合成,从而完成光能向生物能的转化。aR4和bR具有相似的光循环过程,但质子传递时序不同,aR4是先从胞内吸收质子再将质子释放到胞外,而bR恰好相反。甲硫氨酸-145是位于bR视黄醛发色团键合区的一个重要残基,对其光循环有着重要的影响,而在aR4中处在相应位置上的苯丙氨酸-146是其视黄醛键合区与bR唯一不相同的残基。因此通过定点突变,采用紫外可见吸收光谱、动力学光谱、质子泵功能检测、低温透射红外光谱等手段对比分析研究M145F和F146M单点突变对bR和aR4光循环造成的影响,有助于深入理解aR4结构与功能的关系。研究结果表明,M145F突变造成了bR光循环L的丢失和质子泵功能的减弱,而F146M突变并未对aR4的光循环造成显著影响,且突变后aR4质子释放时序没有反转,表明该位置上的残基在两个体系中的作用不尽相同。

  • D97N突变对光受体蛋白古紫质4质子泵和能量转换效率的影响

    Abstract:古紫质4(Archaerhodopsin 4, aR4)是新近发现的古生菌Halobacterium species xz515红膜上唯一的光敏视黄醛蛋白,具有和细菌视紫红质(Bacteriorhodopsin, bR)相似的质子泵功能,但在中性pH条件下,其质子释放和摄取顺序与bR相反。针对质子供体天冬氨酸97(Aspartic acid 97, D97)对aR4光循环、特别是对质子释放摄取顺序和菌株ATP生成率的影响,采用基因定点突变技术,构建了aR4的单突变体D97N,以及相对应的bR单突变体D96N。采用紫外-可见吸收光谱和闪光动力学光谱技术初步研究突变对视黄醛键合区、光反应中间态M态和O态、质子泵功能以及菌株ATP生成率的影响。结果表明,D97N突变对视黄醛紫外-可见光吸收波长没有太大影响,但造成M态衰减时间的显著延长、质子泵功能的消失以及菌株ATP的生成率大幅降低。与bR中的D96作用相比,D97对aR4质子功能的影响有所不同,这可能与D97所处的一个更为疏水性的胞外侧环境有关。

  • 环氧粉末涂层在1.5 mol/L NaCl溶液中的失效行为

    Abstract:本文通过电化学阻抗谱(EIS)对两种环氧粉末涂层在1.5 mol/L NaCl溶液中的失效行为进行了研究,利用扫描电子显微镜(SEM)和电子发射能谱仪(EDS)对涂层底部金属表面的腐蚀产物进行了分析,探讨了环氧粉末涂层在1.5 mol/L NaCl溶液中的失效机理。结果表明,在1.5 mol/L NaCl溶液中,氯离子等腐蚀介质能在涂层中形成传输通道渗透到达涂层金属界面,并参与界面的腐蚀反应,腐蚀产物主要为铁的氧化物和铁的氯化物。涂层失效的电化学过程可以分为三个阶段:前期、中期和后期。

  • 扰动溶液中模拟剥离涂层底部Q345钢阴极极化水平

    Abstract:本文利用分段电极测量了阴极保护下矩形模拟剥离涂层底部Q345钢的电位和电流分布,研究了动态溶液中,不同阴极保护电位,不同缝隙宽度以及漏点尺寸条件下,缝内阴极极化水平。结果表明:扰动作用减弱了缝内阴极极化水平,导致相比静态溶液,动态溶液环境中需要更高水平的阴极保护,才能有效抑制缝内金属腐蚀。缝隙宽度和漏点尺寸对扰动改变缝内极化水平的影响明显。

  • 无溶剂环氧防腐涂层在模拟海水冲刷条件下的电化学行为

    Abstract:通过电化学阻抗谱(EIS)对两种环氧粉末涂层和一种无溶剂环氧液体涂层在60℃流速为2m/s、含1%wt石英砂的3.5%wt NaCl 溶液中的失效行为进行了研究,并利用扫描电镜(SEM)观察了冲刷后涂层的表面形貌,利用激光共聚焦显微镜(CLSM)观察了涂层/金属的断面形貌。结果表明,和静态浸泡相同温度介质条件下相比,流动含砂条件明显加速了无溶剂涂层的失效。冲刷条件下,1#和3#无溶剂涂层由于结构比较致密,失效过程主要分为三个阶段:腐蚀性介质在涂层中传输阶段、基体金属腐蚀发生阶段和基体金属腐蚀发展与涂层失效阶段;2#无溶剂涂层内部由于孔隙较多,涂层较快形成贯穿孔,没有观察到失效中期阶段;涂层体系内部存在孔隙是加速涂层失效的一个重要因素。

  • Mo含量对CrMoN复合涂层的组织结构和性能的影响

    Abstract:采用直流反应磁控溅射技术在M2高速钢基片上制备了不同Mo含量的CrMoN复合涂层,研究了Mo含量的变化对CrMoN复合涂层成分、相结构、化学价态、截面形貌、显微硬度和摩擦性能等的影响.结果表明,随着Mo含量的增加,CrMoN复合涂层的相结构先转变为以fcc-CrN相为基础的(Cr,Mo)N置换式固溶体,后转变为以fcc-γ-Mo2N相为主的混合相,当Mo含量为69.3%原子分数时,伴有少量的bcc-Mo相生成;CrMoN复合涂层的显微硬度先增加后降低,在Mo含量为45.4%时具有最高值;当Mo含量大于45.4 %时,在与对磨副摩擦过程中会生成大量的MoO3相,降低了摩擦系数和磨损率.
