  • 基于熵权的黄河流域生态环境演变特征研究

    Abstract: The ecological environment of the Yellow River Basin is systematic and complicated. It is of great significance to study the quantitative evaluation and change characteristics of the Yellow River Basin by providing scientific basis on integrated river basin management. This in one of the current widespread concerns and urgent problems to be solved. Through calculating the entropy weights of eco-environmental indicators in Yellow River Basin from 1980 to 2019, the dynamic entropy weight results were verified. Then the importance of the individual indicator in system was proved to be variable. On this basis, the EDIs (Environment Development Indexes) of the Yellow River Basin from 1980 to 2019 were analyzed and plotted by curve, which valued between 56.64~76.60. The changing trend of EDI in the Yellow River Basin was almost presented stabilized before 2003, and continuously increasing from 2003. The results show that through the years of protection and restoration, the ecological negative feedback was exerted profound effect, resulting the ecological environment of the Yellow River Basin was changing well.

  • 基于Canopy聚类的噪声自适应模糊C-均值算法

    Abstract: Aiming at the problem that the spatial influence factors are easily misidentified by noise in the fuzzy C-means algorithm (WFLICM) for local spatial information, this paper proposed a fuzzy C-means clustering algorithm for image segmentation (NLWFLICM) based on local and non-local spatial information. It introduced the non-local spatial information into the fuzzy influencing factor of WFLICM algorithm, the weight of local and non-local information is adaptively set according to the noise level, and the fuzzy influence factors of the central point are re-marked. The experimental results show that the NLWFLICM algorithm is more robust and adaptive than the WFLICM algorithm, and improves the robustness of the WFLICM algorithm to a large extent, while preserving the image texture. In order to improve the clustering performance and convergence speed of the algorithm, combined with the advantages of Canopy algorithm for fast clustering of data, this paper proposes an improved algorithm for FCM image segmentation based on Canopy clustering and non-local spatial information (Canopy-NLWFLICM) before clustering algorithm.   • 适用于入侵害虫治理的遗传调控技术

    Abstract:“生物入侵”是指某种生物从外地自然传入或人为引种后成为野生状态,并对本地生态系统造成一定危害的现象。在缺乏天敌和其他制约因子存在的前提下,入侵种能够快速繁殖和扩增并对当地的生态环境、生物多样性和经济造成严重的损失。随着全球化进程的发展,生物入侵变得更加普遍并且危害更加严重,加强入侵种的治理至关重要。由于传统的物理和化学方法在入侵种防治中存在着一些难以解决的问题,开发和利用无污染、无公害的遗传调控技术在生物入侵种的综合治理方面就显得非常迫切。文章针对我国入侵害虫的现状、主要入侵方式及目前的防治措施进行了总结,同时对遗传调控技术及其在生物入侵种综合治理方面的应用进行了阐述,以期为我国生物入侵的科学治理提供参考。

  • 利用CpG DNA甲基化酶M.Sss I共表达载体制备限制性内切酶Not I

    Abstract:限制性核酸内切酶是DNA重组的重要分子生物学工具。由于其本身对DNA有切割作用导致其重组表达在技术上十分困难,产率低、提纯程序复杂。而商业化生产所采用的利用专一型甲基化酶保护宿主DNA的限制酶表达技术流程繁琐、实用性有限。本研究为表达Not I限制酶,采用来源于Spiroplasma sp. MQ1的DNA甲基化酶M.Sss I特异性甲基化CpG序列,甲基化后的DNA会免受识别位点中包含CpG序列的限制酶Not I的切割。将甲基化酶M.Sss I导入大肠杆菌表达宿主ER2566中后,M.Sss I基因在宿主中持续表达并甲基化宿主DNA成CpG甲基化样式;利用此表达体系制备限制性内切酶Not I获得成功。并借助引入纯化标签经过简便的Ni亲和层析和阴离子交换层析2步层析洗脱纯化,制备了高活力高纯度的重组限制酶Not I。此表达体系可应用于一系列识别位点中包含CpG序列的限制酶的表达。
